[Introduction]

This Nucleic Acid Releaser can destroy the protein structure of the virus to release the viral nucleic acids, at the same time it can effectively inhibit the activity of nucleases. It’s suitable for lysis and release of viral nucleic acids (DNA and RNA) from oral and nasopharyngeal swabs. It is also suitable for direct adding and testing of extraction-free nucleic acid amplification (PCR/RT-PCR) reagents due to its good compatibility with amplification reactions. This product is not suitable for preservation of live viruses.

[Protocol]

1. Take out the Nucleic Acid Releaser and invert upside down to mix well to re-suspend the precipitated insoluble particles. Use a 1-mL pipette to pipette 1-2 mL of the Nucleic Acid Releaser into a new centrifuge tube immediately;

2. Put the sampled nasopharyngeal/oral swab into 1-2 mL of the Nucleic Acid Releaser, squeeze and stir repeatedly and finally take out and discard the swab;

3. Heat the centrifuge tube at 98°C for 10mins and pay attention to prevent the cap from bursting during the heating process;

4. Centrifuge at 12000 rpm for 1min;

5. After cooling to room temperature, the supernatant can be directly used as PCR reaction templates. The recommended amount of templates added is 10-20% of the total reaction volume.
